Extracellular vesicle microRNA cargoes from intermittent hypoxia-exposed cardiomyocytes and their effect on endothelium

چکیده

Intermittent hypoxia (IH), a main characteristic of obstructive sleep apnea (OSA) syndrome, is an independent risk factor cardiovascular complications. However, the mechanism has not been fully elucidated. Growing evidence revealed alterations extracellular vesicle (EV) contents, mostly miRNAs, playing pathogenic role in In current study, we attempt to compare disparity myocardial EV miRNA components after IH or normoxia treatment and determine whether EVs from IH-treated cardiomyocytes could affect endothelial function. 63 differentially expressed miRNAs were identified IH-exposed by chip assay. Among them, 16 with homologous sequence mouse human verified qPCR assay 11 proved same tendency as KEGG predicted that function was enriched Akt signaling pathway. Notably, dramatically impaired endothelial-dependent relaxation inhibited Akt/eNOS expression cells. This study provides first significantly alters composition reveals novel under status, which will help understand OSA- IH-related dysfunction new scope. • are validated different EVs. signal pathway miRNAs. impair inhibit expression.

Extracellular Vesicle MicroRNA Transfer in Lung Diseases

MicroRNAs (miRNAs) are single-stranded, small non-coding RNAs that ate involved in the transcriptional and post-transcriptional regulation of gene expression. Recently, miRNAs were demonstrated to be effectively delivered to a target cell or tissue from a host cell via extracellular vesicles (EVs). These EVs can be detected in blood, urine, exhaled breath condensates, bronchoalveolar lavage flu...

the effect of extracellular matrix on choronotropic characteristics of cardiomyocytes drived from embryonic stem cells

introduction: extracellular matrix (ecm) components significantly influence the growth characteristics of cell, development of spontaneous contractile activity, andmorphologic differentiation. the present study evaluates the effect of ecm ongenotypic and physiologic characteristics of cardiomyocytes derived from embryonic stem (es) cells. material and methods: embryoid bodies derived from mouse...

Knockdown of MicroRNA-122 Protects H9c2 Cardiomyocytes from Hypoxia-Induced Apoptosis and Promotes Autophagy

BACKGROUND Acute myocardial infarction (AMI) is a severe disease causing heart failure and sudden death. Studies indicate that microRNAs (miRNAs) are involved in the pathophysiology of AMI. In the present study, we carefully explored the effects of miR-122 on myocardial hypoxia injury and its possible underlying mechanism. MATERIAL AND METHODS miR-122 expression was analyzed in H9c2 cardiomyocy...

Effect of intermittent hypoxia on osteoprotegerin expression in rat liver

Hypoxia is one of the most frequently encountered stress factors in both health and disease. Intermittent hypoxia (IH) is the most common pattern of hypoxic exposure. Osteoprotegerin (OPG) is produced by various cell types, including hepatocytes. Alterations in serum and tissue levels of OPG have been linked to many types of diseases, and clinical significance of OPG expression status has recen...

Intermittent Hypoxia Effect on Osteoclastogenesis Stimulated by Neuroblastoma Cells

BACKGROUND Neuroblastoma is the most common extracranial pediatric solid tumor. Intermittent hypoxia, which is characterized by cyclic periods of hypoxia and reoxygenation, has been shown to positively modulate tumor development and thereby induce tumor growth, angiogenic processes, and metastasis. Bone is one of the target organs of metastasis in advanced neuroblastoma Neuroblastoma cells prod...
